Development of Rat Corneal Transplantation Model and Separate Corneal Laver Transplantation Using this Model

Abstract
An orthotopic penetrating keratoplasty model was developed in the rat. An oversized (0.5 mm) graft was used and interrupted sutures were applied. These sutures were not removed. All grafts (13 grafts) remained clear in the syngeneic group (Lewis rat to Lewis rat transplantation). Eleven grafts out of 13 were rejected by the third week in the disparate group (Brown Norway rat to Lewis rat transplantation).
T suppressor/cytotoxic cells were mainly identified in the rejected cornea.
Human corneal endothelial graft (separate endothelial layer transplantation) was perfornqed in this rat keratoplasty model. All grafts (9 grafts) without immunosuppression were rejected by the 3rd week. However, 4 grafts out of 12, in which cyclosporin was used for immunosuppression, remained clear by the third week. Immunohistochemical study of clear grafts showed that approximately a half of endothelium was transplanted human endothelium. This findings imply that endothelial transplantation only can elicit rejecton reaction in rat keratoplasty model, and that if rejection reaction is prevented by immunosuppression, transplanted endothelium can survise and function normaly.
